Immunofluorescence staining of SIN3A in HeLa cells. Cells were fixed with 4% PFA, permeabilzed with 0.1% Triton X-100 in PBS, blocked with 10% serum, and incubated with SIN3A Polyclonal Antibody at 4°C overnight. Then cells were stained with the Alexa Fluor®488-conjugated Goat Anti-rabbit IgG secondary antibody (green) and counterstained with DAPI (blue). Positive staining was localized to nucleus.

Mammalian Sin3 (mSin3) is a 150 kDa protein closely related to the yeast SIN3 repressor protein involved in the transcriptional repression of many genes. There are two mammalian isoforms, mSin3A and mSin3B. Containing 4 paired amphipathic helix domains (PAH domains), mSin3A and mSin3B have been shown to directly interact with several other transcriptional repressor proteins including HDAC1, HDAC2, RbAp46, the methyl CpG binding protein MeCP2, the Mad/Max heterodimer, and the corepressors silencing mediator of retinoic acid and thyroid hormone receptor (SMRT) and nuclear receptor corepressor (N-CoR).
